What Is ARA-290?

ARA-290, also called cibinetide or pyroglutamate helix-B surface peptide, is an 11-amino-acid synthetic peptide modeled on the outward-facing region of erythropoietin helix B. It was designed to study non-erythropoietic signaling associated with an EPOR/CD131 receptor complex.

Overview

Researchers use ARA-290 to separate stress-response signaling from the classical erythropoietin-receptor pathway associated with red-blood-cell production. Cell and animal models have examined receptor dependence, inflammatory challenge responses, endothelial behavior, and sensory-neuron readouts.

In plain language, ARA-290 helps researchers ask whether one small part of erythropoietin can trigger a cell-stress response without using the main red-blood-cell signaling route.

History

ARA-290 emerged from efforts to map erythropoietin regions linked to tissue-response signaling without retaining the full hormone's canonical hematopoietic activity. The helix-B surface was reduced to a short peptide so that the proposed alternate receptor pathway could be tested directly.

Research Findings

  • Biochemical records identify ARA-290/cibinetide as an 11-residue erythropoietin-derived peptide.
  • Cell studies report binding and signaling associated with EPOR/CD131 rather than the canonical EPOR homodimer.
  • Stress and inflammatory challenges can change CD131 surface expression and downstream transcriptional responses in experimental systems.
  • Observed effects vary by cell type and injury model, and receptor dependence should be confirmed with blocking or genetic controls.
