What Is IGF-1LR3?
IGF-1 LR3 is an engineered 83-amino-acid analogue of insulin-like growth factor 1. It contains an Arg substitution at position 3 and a 13-amino-acid extension at the N-terminus, changes designed to reduce binding to several IGF-binding proteins while retaining IGF-1 receptor activity.
Overview
Researchers use IGF-1 LR3 to study IGF-1 receptor signaling when binding-protein control is reduced relative to native IGF-1. Cell systems can measure receptor phosphorylation, PI3K/AKT and MAPK signaling, proliferation, survival, differentiation, and culture productivity. In plain language, it is a longer-acting laboratory probe for separating receptor signaling from some of the proteins that normally hold IGF-1 in check.
Researchers are interested in IGF-1 LR3 because it helps them study the IGF-1 receptor with less interference from proteins that normally bind and regulate native IGF-1.
History
Long R3 IGF-1 was engineered as a research and cell-culture analogue of native IGF-1. The Arg3 change and N-terminal extension created lower affinity for IGF-binding proteins and a distinct activity profile in vitro. It became a common laboratory reagent in IGF signaling and serum-free culture research.
Research Findings
- Biochemical studies show reduced interaction with several IGF-binding proteins compared with native IGF-1.
- Cell assays measure IGF-1 receptor phosphorylation and downstream PI3K/AKT and MAPK pathway activation.
- Controlled culture studies track proliferation, survival, differentiation, apoptosis, or recombinant-cell productivity.
- Because the analogue can produce a more persistent in-vitro signal, concentration, exposure time, and receptor expression require careful control.
