What Is MT-1 (Melanotan 1)?

Melanotan I, commonly associated with the alpha-MSH analogue afamelanotide, is a synthetic 13-amino-acid melanocortin peptide. Its Nle4 and D-Phe7 substitutions make it more stable than native alpha-MSH and support research focused primarily on MC1 receptor signaling.

Overview

Researchers use MT-1 in melanocyte and receptor models to study the cAMP pathway, pigment-related enzymes, oxidative-stress responses, and gene regulation downstream of MC1R. Compared with broader melanocortin ligands, it is often used when the research question centers on pigment-cell signaling and receptor-dependent cellular responses.

Researchers are interested in MT-1 because it offers a relatively focused way to study how MC1 receptor signaling changes pigment-cell behavior and stress-response pathways.

History

The Nle4,D-Phe7 alpha-MSH analogue was developed by University of Arizona researchers, including Victor Hruby and Mac Hadley, during melanocortin studies in the 1980s. The substitutions were designed to resist rapid breakdown while retaining melanocortin activity. The related pharmaceutical name does not replace the need to verify the identity and specifications of each research lot.
